Klassen des NAWI-Zweiges verbrachten am 4. April einen Vormittag im Vienna Open Lab, einem Mitmachlabor am Zentrum f\u00fcr Molekulare Biologie der Universit\u00e4t Wien. In diesem Labor-Praktikum galt es herauszufinden, ob die Sch\u00fcler*innen das Gen f\u00fcr Langschl\u00e4fer oder das Gen f\u00fcr Kurzschl\u00e4fer in sich tragen. Daf\u00fcr\u00a0 konnten die bereits im Unterricht besprochenen molekularen Techniken, wie die PCR-Methode, Gelelektrophorese, Restriktionsenzyme etc.,\u00a0 selbst angewendet werden.<\/p>\n<p><em>\u201eWir haben das Vienna Open Lab besucht und versucht, durch DNA-Proben festzustellen, ob wir genetisch gesehen Kurz- oder Langschl\u00e4fer sind. F\u00fcr den Versuch haben wir uns als Erstes mithilfe eines Wattest\u00e4bchens Mundschleimhautzellen entnommen, um daraus die DNA zu isolieren. Dann haben wir mit der PCR-Methode einen Abschnitt des Clock-Gens (= Langschl\u00e4fer- bzw. Kurzschl\u00e4fer-Gens) vervielf\u00e4ltigt. <\/em><em>Anschlie\u00dfend\u00a0 wurden\u00a0 die\u00a0\u00a0 PCR-Produkte\u00a0\u00a0 mit\u00a0\u00a0 dem \u00a0\u00a0Restriktionsenzym\u00a0\u00a0 Hgal\u00a0\u00a0 verdaut.\u00a0\u00a0 Dieses Enzym erm\u00f6glicht die DNA an speziellen Stellen zu schneiden. Ist man Langzeitschl\u00e4fer, kann das Enzym\u00a0\u00a0 schneiden\u00a0\u00a0 und\u00a0\u00a0 es\u00a0\u00a0 entstehen\u00a0\u00a0 zwei\u00a0\u00a0 Fragmente.\u00a0\u00a0 Ist\u00a0\u00a0 man\u00a0\u00a0 Kurzzeitschl\u00e4fer, erkennt\u00a0\u00a0 das\u00a0\u00a0 Enzym\u00a0\u00a0 die\u00a0\u00a0 Sequenz\u00a0\u00a0 nicht\u00a0\u00a0 und\u00a0\u00a0 das\u00a0\u00a0 PCR-Produkt\u00a0\u00a0 wird\u00a0\u00a0 nicht\u00a0\u00a0 geschnitten. Schlie\u00dflich konnten wir durch die Durchf\u00fchrung der Gelelektrophorese das Ergebnis deutlich machen.\u201c (Sude und Sila, 8B) <\/em><\/p>\n<p>&nbsp;<\/p>\n<p>Bei der Laborarbeit wurde auch erkannt, dass die kleinsten Ungenauigkeiten in der Durchf\u00fchrung zu Fehlern in den Ergebnissen f\u00fchren k\u00f6nnen. Wer nun Langschl\u00e4fer oder Kurzschl\u00e4fer ist \u2013 und sich damit in Zukunft zumindest teilweise auf seine Gene ausreden kann ? \u00a0\u2013\u00a0 wird noch mithilfe des
